      Applications
      Key Applications Immunoblotting (Western Blotting)
      Immunohistochemistry
      Application NotesImmunoblotting (1:1000)
Immunohistochemistry (see comments; heat pre-treatment required)
Application CommentsAlthough the antibody was adsorbed against a phospho-peptide corresponding to Pyk2, some cross-reactivity may still be detected in cases where Pyk2 is overexpressed relative to FAK within the same cell system. This antibody has also been reported to work for immunohistochemistry; heat pre-treatment with citrate buffer, pH 6.0 is required. When diluted 1:1000 there is sufficient antibody for 10 miniblots at 10 ml per blot. Antibody should be titrated for optimal results in individual systems.

DescriptionRabbit polyclonal antibody adsorbed against non-phosphopeptide corresponding to the immunogen phosphorylation site, followed by immunoaffinity chromatography. Recognizes the ~125 kDa FAK protein phosphorylated on Tyr397.
BackgroundDiscovered as a substrate for Src, FAK acts as a key element of integrin signaling. FAK is a non-receptor tyrosine kinase that plays a central role in cell spreading, differentiation, migration, cell death, and acceleration of the G1 to S phase transition of the cell cycle. The Tyr397 autophosphorylation site binds Src family SH2 and the p85 subunit of PI3-Kinase.
