      BackgroundATM (ataxia telangiectasia mutated, ACC# NP_000042) is a member of the PI-3/PI-4 kinase family and is a central mediator of the cellular response to DNA damage and cellular stress. Mutations in this gene are associated with ataxia telangiectasia, an autosomal recessive disorder. ATM is a regulator of numerous downstream proteins, including the tumor suppressor proteins, p53 and BRCA1, the checkpoint kinase, CHK2, the checkpoint proteins RAD17 and RAD9, and the DNA repair protein NBS1. ATM and the functionally related ATR kinase are thought to be master regulators of cell cycle checkpoint signaling pathways that are required for the cellular response to DNA damage and as well as genome stability. Activation of ATM in response to DNA damage requires intermolecular autophosphorylation of Ser1981 and dimer dissociation.
