      Background InformationHigh affinity nerve growth factor receptor (EC 2.7.10.1; UniProt P35739; also known as Neurotrophic tyrosine kinase receptor type 1, p140-TrkA, Slow nerve growth factor receptor, Trk-A, TrkA neurotrophin receptor, TrkA proto-oncogene receptor, Tropomyosin-related kinase A) is encoded by the Ntrk1 gene (also known as Trk, Trka) in rat (Gene ID 59109). Neurotrophins mediate a wide range of neuronal functions, including survival, differentiation, growth, and death, via two classes of transmembrane receptors, the Trk receptor tyrosine kinase and the p75 neurotrophin receptor. While nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3), and NT-4 all bind the p75 receptor, TrkA is selective for NGF, TrkB is selective for BDNF and NT-4, and TrkC is selective for NT-3. TrkA and TrkB receptors can also be activated by ligands of the G-protein-coupled receptor (GPCRs), including the nucleoside adenosine or adenosine agonists, such as CGS 21680 and the pituitary adenylate cyclase-activating polypeptide (PACAP). In addition to transactivation via GPCRs, Glucocorticoids (GCs) are also reported to promote TrkA/B/C activation due to signaling cross-talk between Trk and GC receptor (GR).
