DescriptionPurified mouse monoclonal antibody generated by immunizing mice with the specified immunogen and fusing splenocytes with NS1 mouse myeloma cells. Recognizes the ~53 kDa wild-type and mutant forms of p53.
Backgroundp53 (393 amino acids) exhibits sequence-specific DNA binding, directly interacts with various cellular and viral proteins, and induces cell cycle arrest in response to DNA damage by inducing GADD45, WAF1, and MDM2. p53 is the most commonly mutated gene in human cancer with most mutations occurring in the DNA contact residues. p53 binds damaged DNA non-specifically via its C-terminal 75 amino acids which in turn may be responsible for subsequent activation, sequence-specific DNA binding, and WAF1 induction allowing DNA repair to take place. p53 down regulates bcl-2 expression and up regulates bax expression, but may not always be necessary for apoptosis. In addition to the afore mentioned proteins, two other proteins involved in replication and recombination, RPA and RAD51, also bind p53.
