DescriptionAnti-Estrogen Receptor α Antibody, clone 60C, rabbit monoclonal
      Alternate Names
      • Estradiol receptor
      • Nuclear receptor subfamily 3 group A member 1
      • estrogen receptor 1
      • estrogen receptor alpha
      • oestrogen receptor
      • steroid hormone receptor
Background InformationThe Estrogen Receptors (ER) belong to the nuclear receptor family that contains a ligand-binding domain that maintains the protein in an inactive conformation unless bound to the hormone. Two ERs are known, ERα, classically known as ER, and ERβ, described long after ERα. Activated Estrogen Receptor is a DNA-binding protein capable of inducing many changes in gene expression. The Estrogen Receptor is believed to play an important role in the growth and development of a subset of hormone dependent human breast cancers. ERβ is expressed in prostate, ovary, and other reproductive organs. Although its steroid specificity is very similar to that of the ERα, the molecular mechanisms regulating the transcriptional activity of ERβ may be distinct from those regulating ERα.

ApplicationAnti-Estrogen Receptor α Antibody, clone 60C is an antibody against Estrogen Receptor α for use in WB & IP.
      Key Applications
      • Western Blotting
      • Immunoprecipitation
      Application NotesImmunoprecipitation:
4-10 μL of a previous lot of this clone immunoprecipitated Estrogen Receptor α from 1 mg of MCF-7 RIPA lysate.

      Biological Information
ImmunogenKLH-conjugated, synthetic peptide corresponding to amino acids 13-32 (C-ALLHQIQGNELEPLNRPQLK) of human Estrogen Receptor α. The immunizing sequence is identical in rat and mouse and has 19/20 identical amino acids in bovine and porcine.

      Entrez Gene Number
Entrez Gene SummarySummary: This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for
hormone binding, DNA binding, and activation of transcription. The
protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast
cancer, endometrial cancer, and osteoporosis. Alternative splicing results in several transcript variants, which differ in their 5' UTRs and use different promoters. [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P03372 # Nuclear hormone receptor. The steroid hormones and their receptors are involved in the regulation of eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues.
SIZE: 595 amino acids; 66216 Da
SUBUNIT: Binds DNA as a homodimer. Can form a heterodimer with ESR2. Interacts with NCOA3, NCOA5 and NCOA6 coactivators, leading to a strong increase of transcription of target genes. Interacts with NCOA7 in a ligand-inducible manner. Interacts with PHB2, PELP1 and UBE1C. Interacts with AKAP13. Interacts with CUEDC2. Interacts with JARID1A. Interacts with SMARD1. Interacts with SLC30A9 (By similarity). Interacts with HEXIM1 and MAP1S. Interacts with PBXIP1.
      SUBCELLULAR LOCATION: Nucleus.
      DOMAIN: SwissProt: P03372 Composed of three domains: a modulating N-terminal domain, a DNA-binding domain and a C-terminal steroid-binding domain.
PTM: Phosphorylated by cyclin A/CDK2. Phosphorylation probably enhances transcriptional activity.
SIMILARITY: Belongs to the nuclear hormone receptor family. NR3 subfamily. & Contains 1 nuclear receptor DNA-binding domain.
MISCELLANEOUS: In the absence of ligand, steroid hormone receptors are thought to be weakly associated with nuclear components; hormone binding greatly increases receptor affinity. The hormone- receptor complex appears to recognize discrete DNA sequences upstream of transcriptional start sites.
