      • Latent membrane protein
      • p63
Background InformationLatent membrane protein 1 (UniProt: P03230; also known as LMP-1, p63) Is encoded by the LMP1 (also known as BNLF1) gene (Gene ID: 3783750) in Epstein-Barr virus. Epstein-Barr virus (EBV) is shown to the causative agent of infectious mononucleosis and it has been associated with human malignancies, such as the African Burkitt's lymphoma and nasopharyngeal carcinoma. EBV is reported to have a unique tropism for resting B lymphocytes and causes them to transform into continuously proliferating lymphoblasts. Latent Membrane Protein 1 (LMP-1) is the dominant oncogene expressed by EBV. It is expressed in the plasma membrane of transformed B cells latently infected with the virus. It is not found in uninfected B cells. It is a type III multi-pass integral membrane protein with short hydrophilic N terminus, six hydrophobic membrane spanning regions, and a relatively long C-terminal cytoplasmic tail. It is only found in cells with EBV genetic information and plays a role in the EBV-induced B-cell transformation and immortalization. It acts as a CD40 functional homolog to prevent apoptosis of infected B-lymphocytes and drive their proliferation. It acts as a constitutively activated receptor of the tumor necrosis factor (TNF) receptor superfamily and activates mitogen-activated kinases, NF-kB, and the JAK/STAT pathway via TNF receptor-associated factors (TRAFs).
