      HostRabbit
SpecificityApoptosis is induced by certain cytokines including TNF and Fas ligand in the TNF family through their death domain containing receptors. TRAIL/Apo2L is a new member of the TNF family. DR4 was recently identified as the receptor for TRAIL. A novel death domain containing receptor for TRAIL was more recently identified and designated DR5, Apo2, TRAIL-R2, TRICK2 or KILLER by several groups independently (Pan et al. 1997; Sheridan et al. 1997; Walczak et al. 1997; MacFarlane et al. 1997; Screaton et al. 1997; Wu et al. 1997; Chaudhary et al. 1997). Like DR4, DR5 transcript is widely expressed in normal tissues and in many types of tumor cells. DR5 binds to TRAIL and mediates TRAIL induced cell death. Overexpression of DR5 induces apoptosis and activates NF-kB.

      Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a member of the TNF-receptor superfamily, and contains an intracelluar death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces apoptosis signal. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: O14763 # Receptor for the cytotoxic ligand TNFSF10/TRAIL. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Promotes the activation of NF- kappa-B.
SIZE: 440 amino acids; 47850 Da
SUBUNIT: Homotrimer. Can interact with TRADD and RIP.
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
TISSUE SPECIFICITY: Widely expressed in adult and fetal tissues; very highly expressed in tumor cell lines such as HeLa S3, K562, HL-60, SW480, A549 and G361; highly expressed in heart, peripheral blood lymphocytes, liver, pancreas, spleen, thymus, prostate, ovary, uterus, placenta, testis, esophagus, stomach and throughout the intestinal tract; not detectable in brain.
DISEASE:SwissProt: O14763 # Defects in TNFRSF10B may be a cause of squamous cell carcinoma of the head and neck (HNSCC) [MIM:275355].
      SIMILARITY: SwissProt: O14763 ## Contains 1 death domain. & Contains 3 TNFR-Cys repeats.
