      Background InformationHigh affinity choline transporter 1 (UniProt: Q9GZV3; also known as Hemicholinium-3-sensitive choline transporter, Solute carrier family 5 member 7, CHT) is encoded by the SLC5A7 (also known as CHT1) gene (Gene ID: 60482) in human. CHT is a multi-pass membrane protein with six extracellular domains, eleven transmembrane domains, and six cytoplasmic domains. It is expressed in cholinergic neurons in putamen, spinal cord, and medulla and serves as a transmembrane transporter that imports choline from the extracellular space into neurons with high affinity in a sodium and chloride ion-dependent manner. In the neurons it is acetylated by the action of choline acetyltransferase to generate acetylcholine. Choline uptake is shown to be the rate-limiting step in acetylcholine synthesis. The majority of CHT is localized intracellularly including a proportion present on acetylcholine-containing synaptic vesicles. Diminished levels of acetylcholine in selected areas of the nervous system are linked to several neurodegenerative diseases. Mutation in SLC5A7 gene have been linked to Neuronopathy and Myasthenic syndrome that is characterized by failure of neuromuscular transmission, fatigability, and muscle atrophy and weakness.
