      Background InformationBeclin 1-associated autophagy-related key regulator (UniProt Q6ZNE5; also known as ATG14 autophagy related 14 homolog, Atg14L, Autophagy-related protein 14-like protein, Barkor, Beclin 1-Interacting protein) is encoded by the ATG14 (also known as ATG14L, BARKOR, KIAA0831) gene (Gene ID 22863) in human. The adaptor protein ATG14/Atg14L/Barkor function as a key driver in orchestrating the formation of autophagosomes, an essential process of autophagy-mediated protein degradation. ATG14 complexes with the class III PI3K catalytic subunit Vps34, the regulatory proteins Beclin 1, and p150 for targeting to the isolation membrane to initiate autophagosomes formation. ATG14 is reported to be negatively regulated by G-protein-coupled receptor (GPCR) signaling, where GPCR-dependent activation of ZBTB16 leads to the degradation of Atg14L and autophagy inhibition.
