DescriptionPlatelet Derived Growth Factor-AA Protein, Recombinant human
OverviewPlatelet Derived Growth Factor (PDGF) is a potent mitogen for a wide range of cell types including fibroblasts, smooth muscle and connective tissue. PDGF, which is composed of a dimer of two chains termed the A chain and B chain, can be present as AA or BB homodimers or as an AB heterodimer. Human PDGF-AA is a 28.5 kDa A chain homodimer protein containing 250 amino acid residues.

      Key Applications
      • Cell Culture
Application NotesFor most in vitro applications, PDGF-AA exerts its biological activity in the concentration range of 0.2 to 10.0 ng/mL. Responding cells are (partial list): Fibroblasts, connective tissue.

Specific ActivityHuman PDGF-AA is fully biologically active when compared to standards, as determined by colorimetric assay using BALB / 3T3 cells.
      Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer or as a heterodimer with the platelet-derived growth factor beta polypeptide, where the dimers are connected by disulfide bonds. Studies using knockout mice have shown cellular defects in oligodendrocytes, alveolar smooth muscle cells, and Leydig cells in the testis; knockout mice die either as embryos or shortly after birth. Two splice variants have been identified for this gene.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P04085 # Platelet-derived growth factor is a potent mitogen for cells of mesenchymal origin. Binding of this growth factor to its affinity receptor elicits a variety of cellular responses. It is released by platelets upon wounding and plays an important role in stimulating adjacent cells to grow and thereby heals the wound.
SIZE: 211 amino acids; 24043 Da
SUBUNIT: Antiparallel disulfide-linked dimer of non-identical (A and B) chains. Homodimers of A and B chains are implicated in transformation processes. Interacts with CSPG4.
      SUBCELLULAR LOCATION: Secreted.
DOMAIN: SwissProt: P04085 The long form contains a basic insert which acts as a cell retention signal.SIMILARITY: Belongs to the PDGF/VEGF growth factor family.
MISCELLANEOUS: A-A and B-B, as well as A-B, dimers can bind to the PDGF receptor.

      Storage and Shipping Information
Storage ConditionsThe lyophilized PDGF-AA is stable for a few weeks at room temperature, but best stored at -80°C. After a quick spin, reconstitute in ddH2O to a concentration of 0.1-1.0 mg/mL. This solution can be diluted into aqueous buffered solutions and stored at 2-8°C for one week or -20°C for future use. Reconstituted PDGF-AA should be stored in undiluted aliquots at -20°C for up to six months.
