Background InformationP90 ribosomal S6 kinase 1 and 2 (RSK1 and RSK2) belong to a family of Ser/Thr kinases that are important for cell cycle, survival, proliferation and other cellular functions. RSK isoforms are made up of two kinase domains and seem to be activated sequentially by a number of phosphorylation events. RSK1 and RSK2 have both been found to be over-expressed in certain cancer types and both could be promising as cancer treatments. RSK2 has recently been implicated in oncogenesis and tumor progression. RSK1 interacts with PKARIalpha through the N-terminal kinase domain and regulates protein kinase A.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a member of the RSK (ribosomal S6 kinase) family of serine/threonine kinases. This kinase contains 2 nonidentical kinase catalytic domains and phosphorylates various substrates, including members of the mitogen-activated kinase (MAPK) signaling pathway. The activity of this protein has been implicated in controlling cell growth and differentiation. Alternate transcriptional splice variants, encoding different isoforms, have been characterized. [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: Serine/threonine kinase that may play a role in mediating the growth-factor and stress induced activation of the transcription factor CREB.

      CATALYTIC ACTIVITY: ATP + a protein = ADP + a phosphoprotein.

      COFACTOR: Magnesium.

ENZYME REGULATION: Activated by multiple phosphorylations on threonine and serine residues.
