      Background InformationDeoxyuridine 5'-triphosphate nucleotidohydrolase (EC 3.6.1.23; also known as dUTPase, dUTP nucleotidohydrolase, or dUTP pyrophosphatase) is encoded by the DUT gene (Entrez Gene ID 1854) in human. dUTPase catalyzes the hydrolysis of dUTP to generate dUMP. This reaction serves to provide dUMP of thymine nucleotides synthesis needed for DNA replication. dUTP hydrolysis by dUTPase also prevents increased incorporation of uracil into DNA, which induces extensive excision repair mediated by uracil glycosylase. This repair process, resulting in the removal and reincorporation of dUTP, is self-defeating and leads to DNA fragmentation and cell death. The DUT gene encodes both mitochondrial and nuclear dATPase (DUT-M and DUT-N, respectively) via alternative splicing. DUT-N (164 aa, 17.7 kDa) corresponds to DUT-M (252 aa, 26.6 kDa) with the first 93 amino acid sequence replaced by MPCSE. Cat. No. MABC608, Anti-dUTPase, clone 6A6.2, is a purified mouse monoclonal antibody. A GST fusion protein containing the full-lenght human DUT-N (UniProt P33316) sequence was used as the immunogen. The binding epitope of clone 6A6.2 is not yet mapped.
