      Background InformationHistone-lysine N-methyltransferase SUV39H1 (UniProt: O43463; also known as EC:2.1.1.355, Histone H3-K9 methyltransferase 1, H3-K9-HMTase 1, Lysine N-methyltransferase 1A, Position-effect variegation 3-9 homolog, Suppressor of variegation 3-9 homolog 1, Su(var)3-9 homolog 1, SUV39H1) is encoded by the SUV39H1 (also known as KMT1A, SUV39H) gene (Gene ID: 6839) in human. SUV39H1 specifically trimethylates lysine 9 of histone H3 using monomethylated H3 lysine 9 as the substrate. It is also reported to weakly methylate histone H1 in vitro. Trimethylation of histone H3 at lysine 9 represents a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. SUV39H1 mainly functions in heterochromatin regions and plays a central role in the establishment of constitutive heterochromatin at pericentric and telomere regions. SUV39H1 is shown to be targeted to histone H3 via its interaction with RB1 and is involved in several key processes, such as repression of MYOD1-stimulated differentiation, regulation of the control switch for exiting the cell cycle and entering differentiation, and regulation of telomere length. It can undergo phosphorylation at serine residues, and to a lesser degree, on threonine residues and the phosphorylated form is stabilized by SBF1 and is less active in its transcriptional repressor function. SUV39H1 contains a SET domain (aa 243-366) that contains the active site, however, both the pre-SET (aa 179-240) and post-SET 9 (aa 396-412) domains are required for the methyltransferase activity.
