      Background InformationHistone H3.1 and H3.2 are closely related histone variants that differ by only one amino acid. Mammalian cells express three major types of noncentromeric histone H3 variants, H3.1, H3.2, and H3.3. H3.1 is only expressed in mammals, while H3.2 is found in all eukaryotes except for yeast. Recent evidence shows that these nearly identical histone proteins differ in their expression profiles and post-translational modifications (PTMs). H3.2 is enriched in modifications associated with transcriptional silencing (di- and tri-methylation of K27), whereas H3.1 is enriched in both K9 dimethylation and K14 acetylation, suggesting that these variants may have separate biological functions.
