      Background InformationT-cell immunoglobulin and mucin domain-containing protein 4 (UniProt Q6U7R4; also known as SMUCKLER, Spleen, mucin-containing, knockout of lymphotoxin protein, T-cell immunoglobulin mucin receptor 4, T-cell membrane protein 4, TIM-4, TIMD-4) is encoded by the Timd4 (also known as Tim4) gene (Gene ID 276891) in murine species. TIM-4 is initially produced with a signal peptide (a.a. 1-22), the removal of which yields the mature type I membrane protein with a large extracellular region (a.a. 23-279), a transmembrane segment (a.a. 280-300), and a small cytoplasmic tail (a.a. 301-343). TIM-4 is expressed by subsets of macrophages, including resident peritoneal macrophages and splenic MOMA-1+-marginal zone macrophages and is shown to bind phosphatidylserine in vitro and support the engulfment of apoptotic cells when expressed in mouse NIH3T3 cells. Milk Fat Globule EGF Factor VIII (MFG-E8) is a soluble protein that also binds phosphatidylserine on apoptotic cells and acts as a bridge between dead cells and phagocytes. MFG-E8 is secreted from a different subset of macrophages and immature dendritic cells than those which express TIM-4. MFG-E8 and Tim4 double knockout mice suffer from autoimmune diseases due to the production of high level of autoantibodies against the large number of unengulfed apoptotic cells.
