Background InformationProgrammed cell death 1 ligand 2 (UniProt: Q9WUL5; also known as PD-1 ligand 2, PD-L2, PDCD1 ligand 2, Programmed death ligand 2, Butyrophilin B7-DC, B7-DC, CD273) is encoded by the Pdcd1lg2 (also known as B7dc, Btdc, Cd273, Pdl2) gene (Gene ID: 58205) in murine species. PD-L2 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 20-221), a transmembrane domain (aa 222-242), and a short cytoplasmic domain (aa 243-247). PD-L2 is a known ligand of PD-1 and is expressed in immature and mature bone marrow-derived dendritic cells and splenic dendritic cells. It is also expressed in placenta and liver and in some tumor cell lines of lymphoid origin. When engaged together with the TCR, the interaction of PD-1 with its ligands delivers an inhibitory signal to T cell proliferation and cytokine production. Higher expression of PD-L2 has been linked to tumor metastasis and prediction of unfavorable prognosis in solid cancer patients. Clone TY25 displays high specificity for PD-L1 and does not react with PD-L1. It is shown to block PD-1 mediated interactions. (Ref.: Yamazaki, T., et al. (2002). J.
