Background InformationKiller cell lectin-like receptor subfamily F member 1 (UniProt: Q9NZS2; also known as Lectin-like receptor F1, Activating coreceptor NKp80, C-type lectin domain family 5 member C) is encoded by the KLRF1 (also known as CLEC5C, ML) gene (Gene ID: 51348) in human. NKp80 is a single-pass type II homodimeric membrane glycoprotein that is strongly expressed in peripheral blood leukocytes and spleen and weaker expression is observed in lymph node and adult liver. It contains a N-terminal cytoplasmic domain (aa 1-38), a transmembrane domain (aa 39-59), and an extracellular domain (aa 60-231). It is reported to be a good marker of NK cell maturity in secondary lymphoid tissues. NKp80 serves as a C-type lectin-like receptor broadly expressed on human NK cells and triggers cytotoxicity via an atypical cytoplasmic hemi-immunoreceptor tyrosine-based activation motif (hemi-ITAM). NKp80 is shown to augment NK cell responses towards malignant and non-malignant myeloid cells. NKp80 is also reported to stimulate the release of proinflammatory cytokines from NK cells and monocytes. Four isoforms of NKp80 have been described that are produced by alternative splicing.
