      Immunoblotting (Western Blotting)
      Paraffin Sections
      Application NotesFrozen Sections (0.06-0.12 µg/ml)
      Immunoblotting (1 µg/ml)
Paraffin Sections (0.06-0.12 µg/ml, heat pre-treatment required, see comments)
Application CommentsStaining of formalin-fixed paraffin sections requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at room temperature for 20 min. Variables associated with assay conditions will dictate the optimal working dilution.

DescriptionProtein G purified mouse monoclonal antibody. Recognizes the ~32-38 kDa α- and β-isoforms of tryptase in mast cells.
BackgroundMast cells are tissue bound cells that normally are distributed throughout connective tissues. They are involved in inflammatory processes as important effector and regulatory cells. Mast cells are known to accumulate at sites of inflammation, such as asthma, allergic rhinitis, and rheumatoid arthritis. Mast cells contain preformed chemical mediators including histamine, chymase, carboxypeptidase and proteolytic tryptase. Mast cells of the skin and lung contain large amounts of tryptase, suggesting this enzyme has a major role in mast cell-mediated events. Human mast cell tryptase has the potential to be used as marker for mast cell activation and as a mediator of inflammation.
