      Background InformationInactive rhomboid protein 1 (UniProt: Q96CC6; also known as iRhom1, Epidermal growth factor receptor-related protein, Rhomboid 5 homolog 1, Rhomboid family member 1, p100hRho, RHBDF1) is encoded by the RHBDF1 (also known as C16orf8, DIST1, IRHOM1) gene (Gene ID: 64285) in human. iRhom1 is a multi-pass, homodimeric, membrane protein that is predominantly localized in the endoplasmic reticulum (ER). It is ubiquitously expressed, but higher expression is observed in brain. It regulates the maturation and activity of ADAM17, a sheddase of the epidermal growth factor (EGF) receptor ligands and TNF . It also controls the transport of ADAM17 from ER to the plasma membrane and removal of its inhibitory propeptide, and stability and degradation. It is reported to play a role in cell survival, proliferation, migration, and inflammation. However, it does not exhibit any protease activity on its own due to lack of amino acids that are required for proteolytic activity. iRhom1-deficient mice are reported to lack mature ADAM17 in the brain.
