Alternate Names
70 kDa zeta-associated protein
Syk-related tyrosine kinase, zeta-chain (TCR) associated protein kinase (70 kD)

Background Information
ZAP70, a 70 kDa member of the Syk tyrosine kinase family, plays a central role in lymphocyte activation and development, and is implicated in several immune disorders. Mutations in ZAP70 gene results in a form of Severe Combined Immunodeficiency Syndrome (SCID) in humans. ZAP70 expression also defines a subset of Chronic Lymphocytic Leukemia (CLL) in patients with unmutated Ig gene and poor clinical course.
Protein G purified mouse IgG2a, Catalog # 05-253, conjugated to fluorescein isothiocyanate (FITC). Note: This product is for research use only. It is not manufacured for diagnostic or therapeutic use in humans or animals.
This gene encodes an enzyme belonging to the protein tyrosine kinase family, and it plays a role in T-cell development and lymphocyte activation. This enzyme, which is phosphorylated on tyrosine residues upon T-cell antigen receptor (TCR) stimulation, functions in the initial step of TCR-mediated signal transduction in combination with the Src family kinases, Lck and Fyn. This enzyme is also essential for thymocyte development. Mutations in this gene cause selective T-cell defect, a severe combined immunodeficiency disease characterized by a selective absence of CD8-positive T-cells. Two transcript variants that encode different isoforms have been found for this gene.
FUNCTION: SwissProt: P43403 # Associates with the T-cell antigen receptor zeta chain (CD3Z). Plays a role in lymphocyte activation. SIZE: 619 amino acids; 69872 Da SUBUNIT: Interacts with SLA2 when it is phosphorylated. Interacts with CD3Z and with phosphorylated NFAM1. Interacts with CBLB (By similarity). Interacts with CBL and SLA when it is phosphorylated. The association with SLA (or SLA2) and CBL probably leads to its destruction. Interacts with SHB. Interacts with DEF6 (By similarity). TISSUE SPECIFICITY: Expressed in T- and natural killer cells. DOMAIN: SwissProt: P43403 The SH2 domain binds to the phosphorylated tyrosine-based activation motif (TAM) of CD3Z. PTM: Phosphorylated on tyrosine residues upon T-cell antigen receptor (TCR) stimulation. Tyr-319 phosphorylation is essential for full activity. DISEASE:SwissProt: P43403 # Defects in ZAP70 are the cause of selective T-cell defect (STD) [MIM:176947]. STD is an autosomal recessive form of severe combined immunodeficiency characterized by a selective absence of CD8-type T-cells. SIMILARITY: Belongs to the protein kinase superfamily. Tyr protein kinase family. SYK/ZAP-70 subfamily. & Contains 1 protein kinase domain. & Contains 2 SH2 domains.

Well-established clinical staging systems continue to form a basis for deciding when to initiate treatment and assigning prognosis in chronic lymphocytic leukaemia. However, these staging systems do not identify those patients with early-stage disease who will progress to require treatment. Recent developments have provided a variety of prognostic markers, which can determine those patients with poor prognosis disease. For example, serum markers (soluble CD23), cell surface (CD38) and cytoplasmic (ZAP70) proteins, cytogenetics and immunoglobulin gene mutational status have all been utilised for this purpose. In order for patients to benefit fully from these discoveries they need to be translated into rapid, standardised and cost-effective clinical laboratory tests. The current range of prognostic markers, and techniques for measuring them are discussed.
Protein tyrosine kinases have long been recognized as the most proximal actors in T-cell antigen receptor (TCR) signaling. Three non-receptor tyrosine kinase families (Src, ZAP-70 and Tec) are known to be critical, but a new study now shows that room needs to be made in this pathway for yet another protein tyrosine kinase family - Abl/Arg.
