Vascular cell adhesion protein 1 (VCAM-1) is a cell surface adhesion molecule belonging to the immunoglobulin cell-cell adhesion receptor family and is comprised of seven Ig-like C2-type domains. VCAM-1 is a crucial component in cell-cell recognition and seems to be involved in cellular adhesion between endothelial cells and leukocytes. It has a regulatory role for both signal transduction and adhesion through its interaction with leukocyte beta-1 integrin VLA4, an interaction that is potentially involved in immune and inflammatory emigration responses of leukocytes. Studies have shown upregulation of VCAM-1 to be critical in responding to inflammatory diseases such as atherosclerosis. VCAM-1 is primarily expressed in endothelial cells and is expressed as two known isoforms found within the cell membrane. Isoform 1 functions as a single-pass type 1 membrane protein, while Isoform 2 acts as a GPI-anchor.

FUNCTION: Important in cell-cell recognition. Appears to function in leukocyte-endothelial cell adhesion. Interacts with the beta-1 integrin VLA4 on leukocytes, and mediates both adhesion and signal transduction. The VCAM1/VLA4 interaction may play a pathophysiologic role both in immune responses and in leukocyte emigration to sites of inflammation.
SUBUNIT STRUCTURE: Binds to ECMV-D capsid proteins and acts as a receptor for this virus.
SUBCELLULAR LOCATION: Isoform 1: Cell membrane; Single-pass type I membrane protein.
TISSUE SPECIFICTY: Expressed on inflamed vascular endothelium, as well as on macrophage-like and dendritic cell types in both normal and inflamed tissue. Expressed in the bone marrow.
PTM: The GPI-anchor is located on position 319 of isoform 2.
Evaluated by Flow Cytometry in mouse bone marrow cells.
Flow Cytometry Analysis: 10 µg of this antibody detected VCAM-1 on mouse bone marrow cells.

Storage Conditions
Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
