Background Information
Histone-lysine N-methyltransferase SETDB2 (UniProt: Q96T68; also known as Chronic lymphocytic leukemia deletion region gene 8 protein, Lysine N-methyltransferase 1F, SET domain bifurcated 2) is encoded by the SETDB2 (also known as C13orf4, CLLD8, KMT1F) gene (Gene ID: 83852) in human. SETDB2 is a member of the class V-like SAM-binding methyltransferase superfamily that is involved in the left-right axis specification in early development and mitosis. Specifically, it trimethylates lysine 9 of histone H3 (H3K9me3). H3K9me3 is a specific tag for epigenetic transcriptional repression that recruits HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones and keeps chromatin in a conformation where the promoter is not accessible for transcription factor binding. SETDB2 is reported to contribute to H3K9me3 in both the interspersed repetitive elements and in centromere-associated repeats and plays a role in chromosome condensation and segregation during mitosis. SETDB2 is shown to be induced by STAT1 in response to type I interferon (IFN /β) signalling to suppress the expression of NF-B target genes to attenuate the inflammatory response. In wound macrophages the expression of SETDB2 is also reported to depend on type I interferon/JAK/STAT1 signaling. SETDB2 has been associated with stimulating cell proliferation, especially in cancer tissue. It is reported to promote hyperproliferation and oncogenesis in acute lymphoblastic leukemia by suppressing CDKN2C activity. (Ref.: Lin, C-H., et al. (2018).
