Background Information
Killer cell immunoglobulin-like receptor 3DL2 (UniProt: P43630; also known as CD158 antigen-like family member K, Natural killer-associated transcript 4, NKAT-4, p70 natural killer cell receptor clone CL-5, p70 NK receptor CL-5, CD158k) is encoded by the KIR3DL2 (also known as CD158K, NKAT4) gene (Gene ID: 3812) in human. KIR3DL2 is a single-pass type I membrane glycoprotein, which is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 22-340), a transmembrane domain (aa 341-360), and a cytoplasmic domain (aa 361-455). It is expressed as a disulfide-linked homodimer and each chain contains three Ig-like domains (42-102; 137-202; and 237-300), and a long cytoplasmic tail that contains two ITIM motifs. Its expression is observed in astrocytes, NK cells, CD8+ T cells, and a small proportion of CD4+ T cells. Its expression is upregulated upon activation of NK and T cells. It serves as a receptor on NK cells and T cells for MHC class I molecules and upon binding of peptide-free HLA-F open conformer, it negatively regulates NK and T cell effector functions. Through interaction with HLA-F, it is suggested to protect motor neurons from astrocyte-induced toxicity. KIR3DL2 also binds to CpG-oligodeoxynucleotides (CpG-ODN), and this ligation induces KIR3DL2 down-modulation from the cell surface and translocation to the endosome to deliver the CpG-ODN to TLR9. Engagement of KIR3DL2 by ODN is reported to induce the internalization of the receptor and caspase-dependent apoptosis of malignant T cells. This apoptotic process is correlated to a dephosphorylation of STAT3 that is constitutively phosphorylated and activated in Se´zary cells. KIR3DL2 is considered as an important diagnostic and prognostic marker for the detection of the tumoral T cell burden of Se´zary syndrome patients. (Ref.: Schmitt, C., et al. (2017). Front.
