      DescriptionAnti-Fractin Antibody, CT
      Alternate Names
      • 32 kDa Fragment of beta-Actin
Background InformationDuring apoptosis, beta actin is cleaved by activated caspase 3 between Asp244 and Gly245. This cleavage results in the release of an approximately 130 amino acid (15 kDa) fragment from the C-terminus of beta actin and a 32 kDa (244 amino acid) N-terminal fragment of actin, known as Fractin. The C-terminus of the Fractin fragment represents an apoptosis-specific neoepitope that is not antigenic in native full-length beta actin and therefore presents a novel and specific marker for distinguishing apoptotic from necrotic cells.

      Applications
ApplicationDetect Fractin using this Anti-Fractin Antibody, C-terminus validated for use in WB, IC, IH(P).
      Key Applications
      • Western Blotting
      • Immunocytochemistry
      • Immunohistochemistry (Paraffin)
Application NotesImmunohistochemistry/Immunocytochemistry: 1:400-1:1000. 1 hr at 37°C or 4°C overnight. Note: If studying paraffin-embedded tissue, the use of antigen retrieval methods is recommended.

Western Blot: 1:1000-1:4000 for incubation overnight at 4°C. Recommended antibody dilution buffer: TBS with 3% BSA and 0.05% Tween 20, pH 7.4

Optimal working dilutions must be determined by end user.
      Biological Information
ImmunogenK-YELPD peptide representing the last five amino acids of the C-terminus of the 32 kDa Fractin fragment of beta-Actin. This sequence is absolutely conserved in Fractin fragments from human, mouse, rat, feline, porcine, ovine, bovine, canine, rabbit and guinea pig origin.
      EpitopeC-terminus
      HostRabbit
SpecificityBy immunohistochemistry and Western blot, this antibody only recognizes the 32 kDa Fractin fragment but not full-length beta actin.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes one of six different actin proteins. Actins are highly conserved proteins that are involved in cell motility, structure, and integrity. This actin is a major constituent of the contractile apparatus and one of the two nonmuscle cytoskeletal actins.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P60709 # Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.
SIZE: 375 amino acids; 41737 Da
SUBUNIT: Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others. Found in a complex with XPO6, Ran, ACTB and PFN1. Component of a complex composed at least of ACTB, AP2M1, AP2A1, AP2A2, MEGF10 and VIM. Interacts with XPO6.
      SUBCELLULAR LOCATION: Cytoplasm, cytoskeleton.
DISEASE: SwissProt: P60709 # Defects in ACTB are a cause of juvenile-onset dystonia [MIM:607371]. Dystonia is a movement disorder with a neurological basis, due to disordered tonicity of muscle. It is characterized by sustained involuntary muscle contractions that cause abnormal postures and repetitive movements. It may affect muscles throughout the body (generalized), in certain parts of the body (segmental), or may be confined to particular muscles or muscle groups (focal). Individuals with juvenile-onset dystonia due to ACTB mutations present a complicated phenotype that includes progressive generalized dopa-unresponsive dystonia, developmental malformations and sensory hearing loss.
SIMILARITY: SwissProt: P60709 ## Belongs to the actin family.
MISCELLANEOUS: In vertebrates 3 main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins coexist in most cell types as components of the cytoskeleton and as mediators of internal cell motility.
