      Background InformationParafibromin (UniProt: Q6P1J9; also known as Cell division cycle protein 73 homolog, Hyperparathyroidism 2 protein, CDC73) is encoded by the CDC73 (also known as C1orf28, HRPT2) gene (Gene ID: 79577) in human. CDC73 is a tumor suppressor that is involved in transcriptional and post-transcriptional control pathways. It is expressed in adrenal and parathyroid glands and in kidney and heart. It is a component of the PAF1 complex with multiple functions during transcription by RNA polymerase II and is implicated in regulation of development and maintenance of embryonic stem cell pluripotency. PAF1 complex associates with RNA polymerase II through the interaction with C-terminal domain. CDC73 is also reported to physically associates with the cleavage and polyadenylation specificity factor (CPSF) and cleavage stimulation factor (CstF) complexes that are required for the maturation of mRNA 3 ends in the cell nucleus. Mutations in CDC73 are reported to cause parathyroid carcinoma and hyperparathyroidism and the loss of CDC73 immunoreactivity is considered as a distinguishing feature of parathyroid carcinoma.
