Anti-APP-C99 Antibody, clone mC99(1-7): SDB (Sicherheitsdatenblätter), Analysenzertifikate und Qualitätszertifikate, Dossiers, Broschüren und andere verfügbare Dokumente.
Amyloid Precursor Protein (APP) is expressed in the brain, kidney, heart, and spleen of fetal tissues; it is induced during neuronal differentiation. The most-substantiated role for APP is in synaptic formation and repair; its expression is upregulated during neuronal differentiation and after neural injury. In adult brain, highest expression of APP is found in the frontal lobe of the cortex. Moderate expression is observed in the cerebellar cortex, the posterior perisylvian cortex-opercular gyri and the temporal associated cortex. Defects in APP are a cause of autosomal dominant Alzheimer's disease (AD). UP on intracerebroventricular injection of this antibody, A beta levels significantly decreased in the brain of APPPS1 mouse model of AD. A related antibody, Cat. No. MABN380, Anti-APP-C99, clone mC99(70-80) was raised against a slightly different region of the same APP-C99 protein.

Selective neutralization of APP-C99 with monoclonal antibodies reduces the production of Alzheimer's Aβ peptides. Houacine, Jemila, et al. Neurobiol. Aging, 33: 2704-14 (2012)
2011
The toxic amyloid-β (Aβ) peptides involved in Alzheimer's disease (AD) are produced after processing of the amyloid precursor protein-C-terminal fragment APP-C99 by γ-secretase. Thus, major therapeutic efforts have been focused on inhibiting the activity of this enzyme. However, preclinical and clinical trials testing γ-secretase inhibitors revealed adverse side effects most likely attributed to impaired processing of the Notch-1 receptor, a γ-secretase substrate critically involved in cell fate decisions. Here we report an innovative approach to selectively target the γ-secretase-mediated processing of APP-C99 with monoclonal antibodies neutralizing this substrate. Generated by immunizing mice with natively folded APP-C99, these antibodies bound N- or C-terminal accessible epitopes of this substrate, and decorated extracellular amyloid deposits in AD brain tissues. In cell-based assays, the same antibodies impaired APP-C99 processing by γ-secretase, and reduced Aβ production. Furthermore, they significantly decreased brain Aβ levels in the APPPS1 mouse model of AD after intracerebroventricular injection. Together, our findings support APP-C99 substrate-targeting antibodies as new immunotherapeutic and Notch-sparing agents to lower the levels of Aβ peptides implicated in AD.
APP is the precursor molecule whose proteolysis generates beta amyloid (Aβ), a 37 to 49 amino acid peptide that is the primary component of amyloid plaques found in Alzheimer's disease patients. Weitere Informationen >>
