      DescriptionInterferon-γ Protein, Recombinant human
OverviewInterferon-g (IFN-gamma) is a lymphoid factor which possesses potent anti-viral activity. It has also been shown to stimulate macrophages and NK cells. Human IFN-gamma is a 16.7 kDa protein containing 143 amino acid residues.

      Applications
ApplicationThe Interferon-g protein (or IFN-gamma protein) is a lymphoid factor which possesses potent anti-viral activity.
      Key Applications
      • Cell Culture
Application NotesFor most in vitro applications, IFN-gamma exerts its biological activity in the concentration range of 0.1 to 10 ng/mL. Responding cells are (partial list): Lymphocytes.
      Biological Information
PurityGreater than 98% by SDS-PAGE and HPLC analyses. Endotoxin level is less than 0.1 ng per μg (1EU/μg) of IFN-gamma.
      SourceHuman, expressed in E.coli
Specific ActivityHuman IFN-gamma is fully biologically active when compared to standards. The ED(50) as determined in a viral resistance assay is <= 0.05 ng/mL corresponding to a specific activity of >= 2 x 10(E7) units/mg.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P01579 # Produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.
SIZE: 166 amino acids; 19348 Da
      SUBUNIT: Homodimer.
      SUBCELLULAR LOCATION: Secreted.
TISSUE SPECIFICITY: Released primarily from activated T lymphocytes.
PTM: Proteolytic processing produces C-terminal heterogeneity, with proteins ending alternatively at Gly-150, Met-157 or Gly-161.
DISEASE: SwissProt: P01579 # In Caucasians, genetic variation in IFNG is associated with the risk of aplastic anemia (AA) [MIM:609135]. AA is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.
SIMILARITY: SwissProt: P01579 ## Belongs to the type II (or gamma) interferon family.

      Storage and Shipping Information
Storage ConditionsThe lyophilized IFN-g, though stable at room temperature, is best stored desiccated at -20°C. After a quick spin, reconstitute in 1x PBS, pH 8.0 to a concentration of 0.5-1.0 mg/mL. Note: Further dilutions should be made into RPMI + 10% FCS or 5% HSA and stored at 4°C for 1 week or -20°C for future use. Reconstituted material should be stored in undiluted aliquots at -20°C for up to six months.
