      Background InformationIntegrin beta 1, also known as CD29, is a 130 kDa transmembrane glycoprotein that forms noncovalent complexes with various Integrin alpha subunits (including alpha 1, alpha 2, alpha 3, alpha 4, alpha 5, and alpha 6) to form the functional receptors that bind to specific extracellular matrix proteins. Integrin receptors are involved in the regulation of a variety of important biological functions, including embryonic development, wound repair, hemostasis, and prevention of programmed cell death. They are also implicated in abnormal pathological states such as tumor directed angiogenesis, tumor cell growth, and metastasis. These heterodimeric receptors bridge the cytoplasmic actin cytoskeleton with proteins present in the extracellular matrix and/or on adjacent cells. Interactions between integrins and the extracellular matrix lead to activation of signal transduction pathways and regulation of gene expression.

      Immunohistochemistry: fresh frozen, or light fixation, with 4% PFA permeabilized with 0.1% triton X-100 in the presence of Mn++ as the epitope of HUTS-4 is dependent upon Mn++ ion. Note antibody only stains activated beta1 integrins. Activated beta-1 integrins are typically found in wound regions for instance (see http://www.ncbi.nlm.nih.gov/books/bv.fcgi?rid=eurekah.figgrp.12951) . Use MAB1981 or CBL1333 for general beta1 integrin staining and detection.

      Western Blotting: non-reducing conditions only (see: http://www.jbc.org/cgi/content/full/271/19/11067).

      SpecificityMonoclonal antibody MAB2079Z is specific for active conformation of human beta1 integrins. The beta1 or VLA integrin subgroup contains at least 10 members, each with a distinct alpha subunit nonconvalently associated with the common beta1 subunit. Functional activation of integrins can be experimentally induced by addition of Mg2+ or Mn2+, or by using one of several stimulatory antibodies that are available. Intracellular signals generated as a result of cell activation also modulate the confirmation and affinity of integrins in cellular systems. Antibodies such as MAB2079Z which have the ability to discriminate between different states of integrin activation are exceptionally useful for studies on the regulation of activation. Antibody MAB2079Z recognizes epitopes in the 355-425 region of the common beta1 subunit of VLA integrins, whose expression parallels the ligand binding activity of these adhesion receptors induced by various extracellular and intracellular stimuli.

Binds LGALS3BP and ITGB1BP3, when associated with alpha-7, but not with alpha-5 (By similarity). Interacts with FLNB and RANBP9. Isoform Beta-1D interacts with ACE2. Isoform Beta-1A interacts with the C-terminal region of FLNC. Interacts with HIV-1 Tat.

      SUBCELLULAR LOCATION: Cell membrane; Single-pass type I membrane protein. Melanosome. Note=Isoform beta-1B does not localize to focal adhesions. Highly enriched in stage I melanosomes.

      TISSUE SPECIFICITY: Isoform beta-1A is widely expressed, other isoforms are generally coexpressed with a more restricted distribution. Isoform beta-1B is expressed in skin, liver, skeletal muscle, cardiac muscle, placenta, umbelical vein endothelial cells, neuroblastoma cells, lymphoma cells, hepatoma cells and astrocytoma cells. Isoform beta-1C and isoform beta-1C-2 are expressed in muscle, kidney, liver, placenta, cervical epithelium, umbilical vein endothelial cells, fibroblast cells, embryonal kidney cells, platelets and several blood cell lines. Isoform beta-C-2, rather than isoform beta-1C, is selectively expressed in primary T-cells. Isoform beta-1C is expressed in nonproliferating and differentiated prostate gland epithelial cells. Isoform beta-1D is expressed specifically in striated muscle (skeletal and cardiac muscle).
