      Brand Family Chemicon®
      Trade Name
      • Chemicon
      DescriptionAnti-Choline Acetyltransferase (ChAT) Antibody
      Alternate Names
      • Choline acetylase
      • acetyl CoA:choline O-acetyltransferase
      • choline acetyltransferase
Background InformationAcetylcholine (ACh) is a common neurotransmitter for motoneurons, preganglionic autonomic neurons, postganglionic parasympathetic neurons, a variety of brain regions and some emerging neuron-like stem cells. The metabolism of Ach is relatively simple, involving only two enzymes: choline acetyltransferase (ChAT) for synthesis and acetylcholinesterase (AChE) for degradation. Further, acetylcholine has little function in neurons other than neurotransmission and seems to be neuron specific. It seems that only cholinergic neurons have significant amounts of ChAT making anti-cholinacetyltransferase a useful specific marker.

      Applications
ApplicationAnti-Choline Acetyltransferase (ChAT) Antibody is an antibody against Choline Acetyltransferase (ChAT) for use in IH, IH(P), IP & WB with more than 70 product citations.
      Key Applications
      • Immunohistochemistry
      • Immunohistochemistry (Paraffin)
      • Immunoprecipitation
      • Western Blotting
      Application NotesImmunoprecipitation:
1:200 dilution of a previous lot immunoprecipitated 68 kDa active ChAT from brain and placenta. Also precipitates a 27 kDa protein that is co-expressed with ChAT (but lacking ChAT activity) in some cholinergic neurons of the central nervous system of Humans and possibly other mammals.

Optimal working dilutions and protocls must be determined by end user.
      Biological Information
      ImmunogenHuman placental enzyme.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes an enzyme which catalyzes the biosynthesis of the neurotransmitter acetylcholine. This gene product is a characteristic feature of cholinergic neurons, and changes in these neurons may explain some of the symptoms of Alzheimer disease. Mutations in this gene are associated with congenital myasthenic syndrome associated with episodic apnea. Multiple transcript variants encoding different isoforms have been found for this gene, and some of these variants have been shown to encode more than one isoform. [provided by RefSeq].

Transcript Variant: This variant (M) contains alternate 5' exon M, uses an upstream start codon, and encodes isoform 2.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P28329 # Catalyzes the reversible synthesis of acetylcholine (ACh) from acetyl CoA and choline at cholinergic synapses.
SIZE: 748 amino acids; 82568 Da
DISEASE: SwissProt: P28329 # Defects in CHAT are the cause of familial infantile myasthenia gravis 2 (FIMG2) [MIM:254210, 254200]; also known as CMS-EA. FIMG2 patients have myasthenic symptoms since birth or early infancy, negative tests for anti-AChR antibodies, and abrupt episodic crises with increased weakness, bulbar paralysis, and apnea precipitated by undue exertion, fever, or excitement. Inheritance is autosomal recessive.
SIMILARITY: SwissProt: P28329 ## Belongs to the carnitine/choline acetyltransferase family.

Quality AssuranceRoutinely evaluated by immunohistochemistry (paraffin) on normal cerebral cortex tissue.

      Immunohistochemistry (Paraffin) Analysis:
ChAT (cat. # AB143) staining on normal cerebral cortex. Tissue treated with no antigen retrieval. The antibody was diluted to 1:100, using IHC-Select® Detection with HRP-DAB. Immunoreactivity is seeing as a staining pattern on the cell body, dendrites and axons of neurons (brown).
