      Background InformationAggregating chondroitin sulfate proteoglycan or aggrecan, is one of the most studied proteoglycans. Many individual monomers of aggrecan bind to hyaluronic acid to form an aggregate, the monomer is known as aggrecan. The aggregates are comprised of up to 100 monomers attached to a single chain of hyaluronic acid. The high content of negatively charged polysaccharide chains contributes to the osmotic swelling of cartilage and in combination with intact collagen fibers results in articular cartilage resistance to compressive forces and high tensile strength. Aggrecan is abundant in articular cartilage and compromises 10% of its dry weight.

The keratan sulfate (KS) and the chondroitin sulfate (CS) attachment domains lie between G2 and G3.

      What is the best primary antibody dilution buffer?At Millipore ,we always recommend to dilute our primary antibody solutions in very simple buffers, usually only TBS or PBS without excess blocking proteins, detergents or other compounds, particularly preservatives such as azide (particularly important if using HRP detections systems: NEVER MIX AZIDE and HRP for the best results as azide is a very powerful inhibitor of HRP]. By keeping the primary antibody simple, this increase antibody-antigen interaction and can increase staining, and when sample has been properly blocked and prepared we have seen increase in background, and often see an improvement in the signal to noise ratio with simple, dilute primary antibody dilution buffers.
