Application CommentsTyr525/526 represent the phosphorylation sites in human Syk, while Tyr519/520 are the equivalent phosphorylation sites in mouse Syk. For immunoblotting the membrane should be incubated with antibody diluted in 5% BSA, 1X TBS, 0.1% Tween®-20 detergent at 4°C overnight with gentle shaking. Antibody should be titrated for optimal results in individual systems.

DescriptionProtein A and immunoaffinity purified rabbit polyclonal antibody. Recognizes the ~72 kDa Syk protein phosphorylated on Tyr525/526.
BackgroundSyk is a protein tyrosine kinase that is widely expressed and plays a key role in hematopoietic cell signaling. Syk is reported to interact with immunoreceptor tyrosine-based activation motifs (ITAMs) found in the cytoplasmic domains of immune receptors. This interaction serves to couple activated immune receptors to downstream signaling events that mediate diverse cellular responses, such as proliferation, differentiation, and phagocytosis. Syk is also reported to act as a potential tumor suppressor in human breast carcinomas. Phosphorylation of Tyr525/526, located in the activation loop of human Syk, is required for Syk activity.
