Detection of human GSK-3β, phosphorylated at Tyr216 by immunoblotting. Samples: Whole cell lysate from control cells (lane 1) or control lysate spiked with human recombinant GSK-3β (lanes 2-5). Primary antibody: PhosphoDetect™ Anti-Glycogen Synthase Kinase-3α/β (pTyr279/216) Rabbit pAb (Cat. No. ST1013) (1:1000) pre-incubated without peptide (lanes 1, 5), or with phosphopeptide immunogen (lane 2), a generic phosphoserine-containing peptide (lane 3), or non-phosphopeptide corresponding to the immunogen (lane 4). Detection: chemiluminescence.

Application CommentsTo generate a positive control, serum starve 3T3-L1 cells for 3 h, then stimulate with 100 nM insulin or 100 ng/ml IGF-1 for 5 min. This antibody has also been reported to work for ELISA and dot blots. At a 1:1000 dilution there is enough antibody for 10 immunoblots at 10 ml/blot. Antibody must be titrated for optimal results in individual systems.

DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~51 kDa phosphorylated GSK-3α protein phosphorylated at Ser279 and the ~47 kDa GSK-3β protein phosphorylated at Ser216.
BackgroundGSK-3α contains a phosphate on tyrosine 279, and GSK-3β contains a phosphate on tyrosine 216. GSK is a protein serine kinase involved in the control of regulatory proteins like glycogen synthase and the transcription factor c-Jun. It has also been associated with the regulation of the microtubule-binding protein Tau, and may be involved in the pathogenesis of Alzheimer's Disease.
