Applications
ApplicationAnti-p73 Antibody, α & β is a Rabbit Polyclonal Antibody for detection of p73 also known as p53-like Transcription Factor & has been validated in IP, WB, IHC(P).
Key Applications
  • Immunoprecipitation
  • Western Blotting
  • Immunohistochemistry (Paraffin)
Application NotesWestern blot: 1 μg/mL of the antibody will detect p73 in 30μg of total cell lysate from overexpressing cell lines. Higher levels of the antibody may be necessary when using native tissue sources.

Immunoprecipitation: Add 1μg of antibody to a sample that has been precleared with Protein A Sepharose™ in 500 μl lysis buffer. Incubate antibody with sample at 4°C for 2 hours with agitation. Follow by incubation with 50 μL Protein A Sepharose™ for 2 hours at 4°C with agitation. Collect the Protein A complex by centrifugation, washing pellet 3 times in lysis buffer. Resuspend final sample in electrophesis buffer, denature by heating at 95°C for 5 minutes, and analyze by SDS-PAGE

Immunohistochemistry of paraffin sections: Signal amplification may be necessary, in conjunction with standard IHC procedures. Dewax and rehydrate sections through descending graded alcohols to water using standard dewaxing protocols. Subject section to heat antigen retrieval. Wash sections 3 times with TBS, pH 7.4. Block endogenous peroxidase activity by incubating the section in 1% H2O2 in methanol for 30 minutes. Wash with TBS and detect according to preferred detection methods. With high sensitivity detection systems, typical working dilutions for the p73 antibody are 1:5,000 - 1;10,000.

Optimal working dilutions must be determined by end user.
Biological Information
ImmunogenGST fusion protein containing aa450-636 of p73 protein.
Epitopealpha and beta
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityAntibody AB7824 recognizes amino acids 450-636 of p73 protein. This region is common to both p73alpha and p73beta isoforms, hence both isoforms of the p73 protein will be detected.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: O15350 # Participates in the apoptotic response to DNA damage. When overproduced, activates transcription from p53-responsive promoters and induces apoptosis. May be a tumor suppressor protein.
COFACTOR: Binds 1 zinc ion per subunit (By similarity).
SIZE: 636 amino acids; 69623 Da
SUBUNIT: The C-terminal oligomerization domain binds to the ABL1 tyrosine kinase SH3 domain. Isoform Beta interacts homotypically and with p53/TP53, whereas isoform Alpha does not. Isoform Gamma interacts homotypically and with all p73 isoforms. Isoform Delta interacts with isoform Gamma, isoform Alpha, and homotypically. Isoforms Alpha and Beta interact with HIPK2. Isoform Alpha interacts with RANBP9. Found in a complex with p53/TP53 and CABLES1. Isoform Beta interacts with WWOX. Interacts with HECW2.
SUBCELLULAR LOCATION: Nucleus.
TISSUE SPECIFICITY: Brain, kidney, placenta, colon, heart, liver, spleen, skeletal muscle, prostate, thymus and pancreas.
DOMAIN: SwissProt: O15350 Possesses an acidic transactivation domain, a central DNA binding domain and a C-terminal oligomerization domain that binds to the ABL tyrosine kinase SH3 domain. & The WW-binding motif mediates interaction with WWOX.
PTM: Isoform alpha (but not isoform beta) is sumoylated on Lys- 627, which potentiates proteasomal degradation but does not affect transcriptional activity.DISEASE:SwissProt: O15350 # Maps to a chromosome region frequently mutated in diverse cell lines of human cancer. Appears not to be frequently mutated in human cancers, in contrast to p53. Hemizygosity is observed in neuroblastoma and oligodendroglioma.
SIMILARITY: Belongs to the p53 family.
MISCELLANEOUS: Activated and stabilized by interaction with RANBP9.
