Background InformationMNTK1 (UniProt: Q76M80; also known as Potassium channel, subfamily K, member 12, Tandem pore domain halothane inhibited K channel, THIK-2) is encoded by the Knck12 (also known as Mntk1) gene (Gene ID: 210741) in murine species. THIK-2 is a member of a novel 2P K1 channel subfamily and is an integral component of plasma membrane with six transmembrane domains. Although it displays about 62% sequence homology with THIK-1, it is not active upon heterologous expression, but is shown to form active heteromeric functional channel along with THIK-1 in the plasma membrane. Conductance of THIK-2 is reported to be smaller ( 2.5 pS), however THIK-1-THIK-2 heterodimers display an intermediate single-channel conductance ( 3.5 pS). The N-terminal region of THIK-2 is reported to be responsible for intracellular retention of the homomeric channel in the endoplasmic reticulum and this retention limits the surface expression of THIK-2 channel. THIK-2 is expressed in several tissues including stomach, liver, and kidney and is particularly abundant in the brain. Murine THIK-2 displays about 98% and 97% sequence homology with human and rat THIK-2, respectively. Murine THIK-2 contains two ion transport domains that are localized in amino acids 111-168 and 224-302. THIK-2 protein has an unusual N terminus containing several repeats of proline, arginine, and cysteine residues. (Ref.: Rajan, S., et al. (2001). J. Biol. Chem. 276(10); 7302-7311; Blin, S., et al.
