Background InformationSerine/threonine-protein kinase Nek2 (EC 2.7.11.1; UniProt P51955; also known as HSPK 21, Never in mitosis A-related kinase 2, NimA-like protein kinase 1, NimA-related protein kinase 2, Protein phosphatase 1 regulatory subunit 111) is encoded by the NEK2 (also known as NEK2A, NLK1, PPP1R111, RP67) gene (Gene ID 4751) in human. Nek2 is a member of the never in mitosis A- (NIMA-) related family of serine/threonine kinases. Nek2 is involved in cell cycle regulation, gene expression, and the maintenance of centrosomal structure and function. Cellular Nek2 levels are tightly regulated throughout the cell cycle with noncentrosomal Nek2 expression peaks from S to G2 phase, while cytoplasmic Nek2 is highest in G1, and nuclear Nek2 is highest during S and G2 phase when it is needed for centrosomal function regulation. During interphase, centrosomes are held together by a proteinaceous linker that connects the the mother and daughter centriole. This linker is disassembled at the onset of mitosis in a process known as centrosome disjunction. The mammalian sterile 20-like kinase 2 (Mst2) and the scaffold protein Salvador (hSav1) are shown to directly interact with Nek2A and regulate its ability to localize to centrosomes, where Nek2A mediating linker dissociation by phosphorylating the centrosomal linker proteins C-Nap1 and rootletin. In addition, Nek2 is also reported to regulate gene expression by phosphorylating the splicing factor SRSF1. Nek2 expression is found upregulated in a wide variety of neoplasms including diffuse large B cell lymphoma and multiple myeloma.
