Background InformationNicastrin (UniProt: Q92542; also known as NCT) is encoded by the NCSTN (also known as KIAA0253, UNQ1874/PRO4317) gene (Gene ID: 23385) in human. Nicastrin is a single-pass type I membrane glycoprotein that is constitutively expressed in neuronal cells. It serves as an essential subunit of the g -secretase complex, an endoprotease complex that catalyzes the intramembrane cleavage of integral membrane proteins such as Notch receptors and amyloid-beta precursor protein. It is reported to bind both full-length b APP and g -secretase substrates (C99- and C83- bAPP fragments) and modulates the activity of g -secretase. In mammalian cells, endogenous as well as over-expressed nicastrin undergoes post-translational N-glycosylation during trafficking from the endoplasmic reticulum to the Golgi apparatus. Presenilin 1 (PS1) is shown to interact preferentially with mature glycosylated form of nicastrin. Inhibition of nicastrin expression is shown to reduce g -secretase activity and its interaction with presenilin1 (PS1). It is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 34-669), a transmembrane domain (aa 670-690), and a short cytoplasmic domain (aa 691-709). Clone A5226A recognizes fully glycosylated mature nicastrin in the active g -secretase complex and is shown to inhibit g -secretase activity by competing with the substrate binding in vitro.
