Background InformationMatrix remodeling-associated protein 8 (UniProt: Q9DBV4; also known as Adipocyte-specific protein 3, Dual Ig domain-containing cell adhesion molecule, DICAM, Limitrin, Mxra8) is encoded by the Mxra8 (also known as Asp3, Dicam) gene (Gene ID: 74761) in murine species. Mxra8 is a single-pass type I membrane protein that is highly expressed at areas of cell-cell contact and may localize to tight junctions. It is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 20-340), a transmembrane domain (aa 341-361), and a cytoplasmic domain (aa 362-442). Its high expression is observed in the brain where it localizes to glia limitans formed by the endfeet of astrocytes surrounding capillaries, and beneath the pia mater. It is also expressed in intercalated disks in the heart and has been detected in pancreatic alpha-cells. It modulates the activity of various signaling pathways via binding to integrin aV:b3.. Mxra8 also acts as an entry mediator for multiple emerging arthritogenic alphaviruses, including chikungunya, Ross River, Mayaro, and O nyong nyong viruses. It is shown to directly bind to chikungunya virus particles and enhances virus attachment and internalization into cells. Clone 9G2.D6 is shown to block chikungunya virus infection in multiple cell types, including primary human synovial fibroblasts, osteoblasts, chondrocytes and skeletal muscle cells.
