Background InformationLeucine-rich repeat-containing protein 4, or LRRC4, also known as Nasopharyngeal carcinoma-associated gene 14 protein or Netrin-G2 ligand (NGL-2), and encoded by the gene LRRC4/BAG, is a synaptic adhesion protein that regulates the formation of excitatory synapses through the recruitment of key pre and post synaptic proteins. LRRC4 is particularly important in auditory synaptic responses and LRRC4 is a key regulator of input-specific synapse development in the hippocampus as well. Finally, LRRC4 appears important in the suppression of glioma cancers in the brain and is specifically down regulated in tumors, and is currently being investigated as a potential anti-cancer target. LRRC4 is localized to the cell junctions and synapses. It is a cell membrane bound, post-synaptic, protein, and LRRC4’s expression is brain specific.
