Background InformationEpidermal growth factor (UniProt: P01133; also known as Urogastrone) is encoded by the EGF gene (Gene ID: 1950) in human. Epidermal growth factor is a single-pass type I membrane protein that is expressed in kidney, salivary gland, cerebrum, and prostate. It stimulates the growth of various epidermal and epithelial tissues in vivo and in vitro and of some fibroblasts in cell culture. It exerts its effects by binding to epidermal growth factor receptor (EGFR) that undergoes homo- or heterodimerization with erbB2 receptors, causing autophosphorylation of its tyrosine residues that in turn activate PI3K, MAPK, or phospholipase C-gamma signaling cascades. EGF is shown to stimulate magnesium reabsorption in the renal distal convoluted tubule via engagement of EGFR and activation of the magnesium channel TRPM6. EGF is synthesized with a signal peptide (aa 1-22) that is subsequently cleaved off to generate the mature form that contains a long extracellular domain (aa 23-1032), a transmembrane domain (aa 1033-1053), and a cytoplasmic domain (aa 1054-1207). Three isoforms of EGF has been described that are produced by alternative splicing. Mutations in EGF gene have been linked to hypomagnesemia that is characterized by massive renal hypomagnesemia and normal levels of serum calcium and calcium excretion, which leads to seizures and mild-to moderate psychomotor retardation.
