Background InformationCMRF-35-like molecule 4(UniProt: Q7TSN2; also known as CLM-4, CD300C molecule 2, Dendritic cell-derived Ig-like receptor 1, DIgR1, Immunoglobulin superfamily member 7, IgSF7, Leukocyte mono-Ig-like receptor 2, Myeloid-associated immunoglobulin-like receptor 2, MAIR-2, MAIR-II) is encoded by the Cd300c2 (also known as Cd300c, Clm4, Igsf7, Lmir2) gene (Gene ID: 140497) in murine species. CLM-4 is a single-pass type I membrane protein that is present on the surface of mast cells, dendritic cells, peritoneal macrophages and a subset of B-cells. It acts as an activating receptor in mast cells and macrophages. It is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 25-187), a transmembrane domain (aa 188-208), and a cytoplasmic domain (aa 209-228). CLM-4 is shown to associate with DAP12, which mediates activating signals, resulting in inflammatory cytokine secretion from macrophages. Cross-linking CLM-4 with this monoclonal antibody is reported to induced secretion of a significant amount of the inflammatory cytokines TNF-alpha and IL-6 from DAP12 -/- as well as wild-type (WT) peritoneal macrophages. Mice deficient in CLM-4 are shown to be more susceptible to caecal ligation and puncture (CLP)-induced peritonitis. (Ref.: Totsuka, N., et al. (2014). Nat. Commun. 5; 4710; Nakahashi, C. et al.
