Description

DescriptionAnti-CDK5RAP2 Antibody
Alternate Names
  • CDK5 regulatory subunit-associated protein 2
  • CDK5 activator-binding protein C48
  • Centrosome-associated protein 215
Background InformationAlso known as Cep215, CDK5RAP2 is involved in a variety of cellular processes. It serves as a potential regulator of CDK5 activity through its interaction with CDK5R1. It acts as a negative regulator of centriole disengagement, which maintains centriole engagement and cohesion. It is involved in the regulation of mitotic spindle orientation. Additionally, this protein mediates spindle checkpoint activation by serving as a transcriptional regulator of the MAD2 and BUBR1 promoter. In conjuction with MAPRE1, CDK5RAP2 promotes microtubule polymerization, bundle formation, and growth at the plus ends. It also plays a role in DNA damage-induced G2 cell cycle checkpoint. CDK5RAP2 is known to be mutated in primary microcephaly, a neuodevelopmental disorder that is characterized by a decrease in brain size.

Entrez Gene Number
Entrez Gene SummaryNeuronal CDC2-like kinase, which is involved in the regulation of neuronal differentiation, is composed of a catalytic subunit, CDK5, and an activating subunit, p25NCK5A. The protein encoded by this gene binds to p25NCK5A and therefore may be involved in neuronal differentiation. The encoded protein may also be a substrate of neuronal CDC2-like kinase. Multiple transcript variants exist for this gene, but the full-length nature of only two has been determined.
Gene Symbol
  • CDK5RAP2
Purification MethodAffinity Purfied
UniProt Number
UniProt SummaryFUNCTION: Potential regulator of CDK5 activity via its interaction with CDK5R1. Negative regulator of centriole disengagement (licensing) which maintains centriole engagement and cohesion. Involved in regulation of mitotic spindle orientation. Plays a role in the spindle checkpoint activation by acting as a transcriptional regulator of both BUBR1 and MAD2 promoter. Together with MAPRE1, it may promote microtubule polymerization, bundle formation, growth and dynamics at the plus ends.

SUBUNIT STRUCTURE: Interacts with CDK5R1 (p35 form). CDK5RAP1, CDK5RAP2 and CDK5RAP3 show competitive binding to CDK5R1. Probably forms a complex with CDK5R1 and CDK5. Interacts with PCNT; the interaction is leading to centrosomal and Golgi localization of CDK5RAP2 and PCNT. Interacts with AKAP9; the interaction is leading to Golgi localization of CDK5RAP2 and AKAP9. Interacts with MAPRE1; the interaction is leading to microtubule attachment at plus ends of CDK5RAP2 and MAPRE1. Interacts with TUBG1; the interaction is leading to the centrosomal localization of CDK5RAP2 and TUBG1. Interacts with TUBGCP3. Interacts with CALM1. Interacts with CDC20.

SUBCELLULAR LOCATION: Cytoplasm > cytoskeleton > centrosome. Golgi apparatus. Cytoplasm. Note: Found in the pericentriolar region adhering to the surface of the centrosome and in the region of the centrosomal appendages. Localizes to microtubule plus ends.

TISSUE SPECIFICITY: Widely expressed. Expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas.
Molecular Weight~220 kDa observed. Uniprot.org states four isoforms produced by alternative splicing, which may be observed at a molecular weight of ~216 kDa, ~211 kDa, ~210 kDa, and ~206 kDa in some cell lysates.
