Background InformationCMRF35-like molecule 8 (UniProt: Q6SJQ0; also known as CLM-8, CD300 antigen-like family member A, Leukocyte mono-Ig-like receptor 1, Mast cell-derived paired immunoglobulin-like receptor 1, Myeloid-associated immunoglobulin-like receptor 1, MAIR-1, MAIR-I, CD300a) is encoded by the Cd300a (also known as Clm8, Lmir1, Mcpir1) gene (Gene ID: 217303) in murine species. CLM-8 is a single-pass type I membrane protein that is present on the surface of most myeloid cells and a subset of B-cells. It has also been shown to be present on the surface of NK cells following IL-12 stimulation. It serves as an inhibitory immunoreceptor and contributes to the down-regulation of cytolytic activity in natural killer (NK) cells, and down-regulates mast cell degranulation. CLM-8 is also reported to negatively regulate the Toll-like receptor (TLR) signaling mediated by MYD88. CLM-8 is shown to deliver its inhibitory signal in mast cells to suppress the production of LPS-induced inflammatory cytokines and chemokines. Its inhibitory signal depends on the phosphorylation of tyrosine residues embedded in Immunoreceptor tyrosine-based inhibitory motif (ITIM) of the cytoplasmic tail. CLM-8 is synthesized with a signal peptide (aa 1-27), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 28-185), a transmembrane domain (aa 186-206), and a cytoplasmic tail (aa 207-318).
