Background InformationTumor necrosis factor receptor superfamily member 11A (UniProt: Q9Y6Q6; also known as Osteoclast differentiation factor receptor, ODFR, Receptor activator of NF-KB, CD265) is encoded by the TNFRSF11A (also known as RANK) gene (Gene ID: 8792) in human. CD265 is a single-pass type I membrane protein that serves a receptor for TNFSF11/RANKL and is considered to be essential for RANKL-mediated osteoclastogenesis. It has ubiquitous expression, but high expression is observed in skeletal muscle, thymus, liver, colon, small intestine and adrenal gland. It is also expressed on dendritic cells and facilitates immune signaling. It interacts with various TRAF family proteins and induces the activation of NF-kappa B and MAPK8/JNK. It is shown to bind to the clefts between the subunits of the TNFSF11/ RANKL ligand trimer to form a heterohexamer. CD265 is synthesized with a signal peptide (aa 1-29), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 30-212), a short transmembrane domain (aa 213-233), and a cytoplasmic domain (aa 234-616). It also contains four extracellular cysteine-rich pseudorepeats.
