Background InformationAtaxin-1 (UniProt: P54253; also known as Spinocerebellar ataxia type 1 protein) is encoded by the ATXN1 (also known as ATX1, SCA1) gene (Gene ID: 6310) in human. Ataxin-1 is a evolutionarily conserved polyglutamine-containing protein that is widely expressed in the normal human brain and peripheral tissues. Within neurons, it is predominantly found in the nucleus, however, some cytoplasmic Ataxin-1 is also found in Purkinje cells and brain stem nuclei. Ataxin-1 is a chromatin-binding factor that represses Notch signaling in the absence of Notch intracellular domain by acting as a CBF1 corepressor. Ataxin-1 can be sumoylated at multiple lysines (K16, K194, K610, K697, and K746) and can undergo phosphorylation at serine 775 that increases its pathogenicity with an expanded polyglutamine tract. Polyglutamine expansion of Ataxin-1 is the cause of spinocerebellar ataxia type 1 that results in progressive loss of motor control. (Ref.: Sanchez, I., et al. (2016). Human Mol.
