Background InformationAnoctamin-6 (UniProt: Q4KMQ2; also known as Small-conductance calcium-activated nonselective cation channel, SCAN channel, Transmembrane protein 16F, TMEM16F) is encoded by the ANO6 (also known as TMEM16F) gene (Gene ID: 196527) in human.TMEM16F is a multi-pass membrane glycoprotein that has 6 cytoplasmic domains, ten transmembrane domains, and four extracellular domains. It is expressed in embryonic stem cell, fetal liver, retina and in chronic myologenous leukemia and intestinal cancer. TMEM16F serves as a small-conductance calcium-activated nonselective cation channel that acts as a regulator of phospholipid scrambling in platelets and osteoblasts. Phospholipid scrambling results in surface exposure of phosphatidylserine which in platelets is shown to be essential to trigger the clotting system. In osteoblasts it is required for the deposition of hydroxyapatite during bone mineralization. It is also shown to generate outwardly rectifying chloride channel currents in airway epithelial cells and Jurkat T lymphocytes. Mutations in ANO6 gene have been linked to Scott syndrome, a mild bleeding disorder due to impaired surface exposure of procoagulant phosphatidylserine on platelets and other blood cells following activation with calcium elevating agents.
