DescriptionAnti-Tyrosine Hydroxylase Antibody
Alternate Names
  • Tyrosine 3-hydroxylase
  • Tyrosine 3-hydroxylase
  • tyrosine hydroxylase
Background InformationTyrosine hydroxylase plays an important role in the physiology of adrenergic neurons. It is the first and rate-limiting enzyme involved in the biosynthesis of the catecholamines Dopamine and Norepinephrine from tyrosine. TH is, therefore, a useful marker for dopaminergic and noradrenergic neurons. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing.

Control
  • Positive: brain (corpus striatum) and adrenal gland. Negative: liver.
PresentationPurified sheep polyclonal in buffer containing 10 mM HEPES, pH 7.5, 150 mM NaCl, 100 μg/mL BSA and 50% glycerol.

Applications
ApplicationAnti-Tyrosine Hydroxylase Antibody is an antibody against Tyrosine Hydroxylase for use in IH & WB.
Key Applications
  • Immunohistochemistry
  • Western Blotting
Application NotesImmunohistochemistry(peroxidase): A previous lot of this antibody was used at 1:1,000 dilution. The antibody gives specific labeling of noradrenergic axons in primate cerebral cortex (Brain Res., 1989, 500:313-324.).
Note: It is important that you use a secondary antibody that has significant reactivity to sheep IgG1 since the majority of the sheep immunoglobulin is IgG1.
Optimal working dilutions must be determined by the end user.
Biological Information
ImmunogenNative tyrosine hydroxylase from rat pheochromocytoma (J. Biol. Chem.,1982, 257:9416-9423).
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostSheep
SpecificityTyrosine Hydroxylase (TH, Tyrosine Monooxygenase). Cross-reacts with all mammalian forms tested to date and some non-mammalian forms.

Entrez Gene Number
Entrez Gene SummaryTyrosine hydroxylase is involved in the conversion of tyrosine to dopamine. As the rate-limiting enzyme in the synthesis of catecholamines, tyrosine hydroxylase has a key role in the physiology of adrenergic neurons. Mutations in this gene have been associated with autosomal recessive Segawa syndrome. Alternatively spliced transcript variants encoding different isoforms have been noted for this gene. [provided by RefSeq].

Transcript Variant: This variant (2) uses a different donor splice site at the first coding exon and is missing an adjacent in-frame coding exon compared to transcript variant 1, resulting in an isoform (b) missing a 31 aa segment compared to isoform a.
Gene Symbol

Purification MethodImmunoAffinity Purified
UniProt Number
UniProt SummaryFUNCTION: SwissProt: P07101 # Plays an important role in the physiology of adrenergic neurons.
COFACTOR: Fe(2+) ion.
SIZE: 528 amino acids; 58524 Da
TISSUE SPECIFICITY: Mainly expressed in the brain and adrenal glands.
DISEASE: SwissProt: P07101 # Defects in TH are the cause of autosomal recessive Segawa syndrome [MIM:605407]; also known as DOPA-responsive dystonia. Typically, it begins in childhood or adolescence with progressive difficulty in walking and, in some cases, spasticity. Some cases present with parkinsonian symptoms in infancy and are referred to as autosomal recessive infantile parkinsonism.
SIMILARITY: SwissProt: P07101 ## Belongs to the biopterin-dependent aromatic amino acid hydroxylase family.

Usage Statement
  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage ConditionsStable for 6 months at -20 to -80°C in undiluted aliquots from date of receipt. Store in a NON-self-defrosting freezer.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
