Background InformationPhosphodiesterase 4D3 (PDE4D3) is a member of the PDE cyclic nucleotides. PDE4D3 is known to be regulated by protein kinase C (PKC)-Raf-MEK-ERK and also by cAMP in vascular smooth muscle cells. PDE4D3 is a direct target of the MSH/cAMP/MITF pathway and may play a role in melanocyte differentiation. PDE4D3 is essential to have equilibrium in relaxing and contracting cues to airway smooth muscle. Down-regulation of PDE4D3 may be associated with analgesic and anti-inflammatory effects. PDE4D3 has also been linked to the risk of stroke, and in particular to polygenic stroke.

TISSUE SPECIFICTY: Widespread; most abundant in skeletal muscle. Isoform 6 is detected in brain. Isoform 8 is detected in brain, placenta, lung and kidney. Isoform 7 is detected in heart and skeletal muscle.

PTM: Isoform 3 and isoform 7 are activated by phosphorylation (in vitro), but not isoform 6. Isoform N3 and isoform 12 are phosphorylated on Ser-49, Ser-51, Ser-55 and Ser-59.

INVOLVEMENT IN DISEASE: Note=Genetic variations in PDE4D might be associated with susceptibility to stroke. Ref.16 states that association with stroke has to be considered with caution.
