Applications
ApplicationAnti-Neurite Outgrowth Inhibitor A/B/C Antibody is an antibody against Neurite Outgrowth Inhibitor A/B/C for use in ELISA & WB.
Key Applications
  • ELISA
  • Western Blotting
Application NotesWestern blot: 1-10 μg/mL using ECL. The calculated molecular weight of Nogo-A is approximately 135 kDa. However a molecular weight of approximately 180 kDa has been reported for full length Nogo-A (Chen et al. 2000; GrandPre et al. 2000; Goldberg & Barres 2000; Prinijha et al. 2000; Tessier-Lavigne & Goodman 2000; Nagase et al. 1998).

ELISA: 1:10,000-1:100,000 using 50-100 ng control peptide per well.

Optimal working dilutions must be determined by the end user.
Biological Information
ImmunogenA 13 amino acid peptide between the putative 2nd-3rd transmembrane domain of human Nogo-A (Chen et al. 2000; GrandPre et al. 2000; Goldberg & Barres 2000; Prinijha et al. 2000; Tessier-Lavigne & Goodman 2000; Nagase et al. 1998).
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityRecognizes human Neurite Outgrowth Inhibitor Protein A/B/C (Nogo-A/B/C). The immunogen peptide is within the 25 amino acid region identified for the inhibition of neurite outgrowth (Chen et al. 2000; GrandPre et al. 2000; Goldberg & Barres 2000; Prinijha et al. 2000; Tessier-Lavigne & Goodman 2000; Nagase et al. 1998).The immunogen shows no significant sequence homology with other known proteins.

SPECIES REACTIVITIES: The immunogen peptide is 92% conserved in rat Nogo or rat foocen-m1.2 protein (Chen et al. 2000; GrandPre et al. 2000; Goldberg & Barres 2000; Prinijha et al. 2000; Tessier-Lavigne & Goodman 2000; Nagase et al. 1998). Reactivity with other species has not been confirmed.

Entrez Gene Number
Entrez Gene SummaryThis gene belongs to the family of reticulon encoding genes. Reticulons are associated with the endoplasmic reticulum, and are involved in neuroendocrine secretion or in membrane trafficking in neuroendocrine cells. The product of this gene is a potent neurite outgrowth inhibitor which may also help block the regeneration of the central nervous system in higher vertebrates. Alternatively spliced transcript variants derived both from differential splicing and differential promoter usage and encoding different isoforms have been identified.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q9NQC3 # Potent neurite growth inhibitor in vitro and plays a role both in the restriction of axonal regeneration after injury and in structural plasticity in the CNS. Isoform 2 reduces the anti-apoptotic activity of Bcl-xl and Bcl-2. This is likely consecutive to their change in subcellular location, from the mitochondria to the endoplasmic reticulum, after binding and sequestration. Isoform 2 and isoform 3 inhibit BACE1 activity and amyloid precursor protein processing.
SIZE: 1192 amino acids; 129931 Da
SUBUNIT: Binds to RTN4R. Interacts with Bcl-xl and Bcl-2. Isoform 2 binds to NGBR and RTN3. Isoform 2 and isoform 3 interact with BACE1 and BACE2.
SUBCELLULAR LOCATION: Endoplasmic reticulum membrane; Multi-pass membrane protein. Note=Anchored to the membrane of the endoplasmic reticulum through 2 putative transmembrane domains.
TISSUE SPECIFICITY: Isoform 1 is specifically expressed in brain and testis and weakly in heart and skeletal muscle. Isoform 2 is widely expressed except for the liver. Isoform 3 is expressed in brain, skeletal muscle and adipocytes. Isoform 4 is testis- specific.
DOMAIN: SwissProt: Q9NQC3 Three regions, residues 59-172, 544-725 and the loop 66 amino acids, between the two transmembrane domains, known as Nogo- 66 loop, appear to be responsible for the inhibitory effect on neurite outgrowth and the spreading of neurons. This Nogo-66 loop, mediates also the binding of RTN4 to its receptor (By similarity).
SIMILARITY: Contains 1 reticulon domain.
