Background InformationN-methylpurine-DNA glycosylase (also known as 3-alkyladenine DNA glycosylase, 3-methyladenine DNA glycosidase, ADPG, DNA-3-methyladenine glycosylase) is encoded by the MPG (also called AAG, ANPG, MID1) gene in human (NCBI Gene ID 4350). MPG/AAG protein is involved in repair of DNA damage due to alkylation both in the nucleus and mitochondria. Specifically, MPG/AAG protein catalyzes the hydrolysis of the N-glycosyl bond between the alkylated base and deoxyribose, thereby removing 3-methyladenine and 7-methylguanine to form "abasic sites" in the damaged DNA. MPG/AAG protein is also found to exhibit DNA-O-glycosidase activity. MPG/AAG protein is reported to remain associated with the abasic DNA after base removal and, in the presence of small alcohols, catalyze the formation of an O-glycosidic linkage between the alcohol hydroxyl group and C1' of the abasic deoxyribose. Unpon alcohol removal, MPG/AAG protein is shown to catalyze the reverse reaction by hydrolyzing the O-glycosidic bonds of alcohol-DNA adducts. (PMID 23290262 & 23688261). Clone 9F7.1 detects an epitope within human MPG/AAG protein (UniProt P29372) N-terminal region, which is not well conserved in murine protein (UniProt Q04841).
