Application NotesInhibition of cell adhesion to fibronectin: 10 μg/mL

Note that where cell adhesion to fibronectin involving other fibronectin receptors, such as VLA-4 (alpha4beta1) integrin, is involved, inhibition of cell adhesion by BMB5 alone may not be complete.

SpecificityAntibody MAB2514 was raised against mouse integrin VLA-5 (alpha5beta1), a fibronectin receptor. The antibody was prepared by fusion of spleen cells from a Fisher rat immunized with mouse embryonal carcinoma P19 cells with myeloma P3X63Ag9.653 cells.Specificity of the antibody was verified by its ability to immunoprecipitate alpha5beta1 heterodimers from (125)-I-surface labeled cells, reciprocal depletion of VLA-5 antigen from cell lysate with antiserum against the cytoplasmic domain of a5 integrin subunit, and immunoprecipitation of VLA-5 integrin from cells known to express this integrin. Cell lines tested to express alpha5beta1, include: P19 cells, mouse melanoma B16F1 cells, mouse squamous carcoma SCC cells and thymocytes. Characterization by immunoprecipitation was typically analyzed by SDS-PAGE using 6% gels under non-reducing conditions.
